We are working on two separate projects, assessing the activation state of Ras in human malignancies and studying the mechanism(s) of nitric oxide (NO) regulation of a vitamin B12 (cobalmin) dependent reaction. We are assessing Ras activation in breast, lung and prostate cancers using a novel enzyme-based method developed in our laboratory. This work is being done in collaboration with Drs. Anna Dreilinger and Anne Wallace (breast cancer), Jim Harrell (lung cancer) and Michael Albo (prostate cancer). We have recently extended this assay to assess the activation states of two other Ras-related proteins, Rap 1 and Rho, and are in the process of using these assays in human tumors. In the second major project, we are studying how NO regulates methionine synthase, and thus carbon flow through the folate pathway.
